Cellular signal transduction occurs via the insulin receptor tyrosine kinase (IRTK), triggering phosphorylation of IRS-1 and IRS-2, which activates the phosphatidylinositol 3-kinase (PI3K)-Akt cascade and downregulates FOXO1 transcription factors. Impaired signaling causes systemic hyperinsulinemia, attenuated mitochondrial beta-oxidation, and accelerated hepatic de novo lipogenesis (DNL).
